Linear DNA migrates through agarose at a rate that is inversely proportional to the log of its molecular weight. You can now generate a standard curve using the DNA molecules in the molecular weight (MW) standard. Measure the distance from the well that each DNA molecule in the MW standard has traveled through the agarose. The molecular weights of each of these fragments are known. Plot the distance migrated by the DNA molecules in the MW standard on the x-axis of your graph and the Log MW on the y-axis.
